The vascular endothelial growth factor (VEGF) is a key-mediator of angiogenesis in physiological and pathological conditions. Under low oxygen conditions (hypoxia) VEGF expression is strongly enhanced via its HIF-1 dependent transcription VEGF mRNA stabilization and internal ribosome entry site (IRES)-mediated translation. Two independent IRESs in the VEGF mRNA 5’-untranslated region confer sustained VEGF mRNA translation under hypoxia when 5'-cap-dependent mRNA translation is reduced. VEGF IRES-mediated initiation of translation requires the modulated interaction of trans-acting factors. An RNA affinity purification approach based on extracts derived from MCF-7 cells cultured under normoxia or hypoxia (1% oxygen) revealed the DEAD-box RNA helicase 6 (DDX6) as VEGF mRNA trans-acting factor. Recombinant DDX6 inhibits VEGF IRES-mediated translation in normoxic MCF-7 cell extracts in vitro while depletion of DDX6 by RNAi promotes VEGF expression in hypoxic MCF-7 cells. |
