This work investigates whether breadcrustextract (BCE) and individual fractions thereof can activate the transcription factor NF-κB. For this purpose, it should be checked whether the activation depends on the expression of the receptor of advanced glycation end products (RAGE). Cardiac mouse fibroblast cellline and three NF-κB reportercelllines were stimulated with BCE and the fractions, respectively. Furthermore, the RAGE receptor population of cell lines was examined. The RAW-cellline shows the highest activation of the NF-κB pathway when stimulated with BCE as well as with the fractions compared to the other cell lines. In addition, this cell line shows the strongest RAGE mRNA band in the Western Blot. The NF-κB signaling pathway of the HEK, Jurkat and RAW celllines were activated by BCE and / or individual fractions of the bread crust. The level of NF-κB activation by BCE or by the fractions used appears to be dependent on receptor loading, but is not statistically significant.
