In der vorliegenden Arbeit wurden neuartige künstliche Bindeproteine entwickelt die auf Ca2+-bindenden Gerüstproteinen der βγ-Kristallin-Familie basieren. Diese fungieren als molekulare Schalter die eine hohe Affinität zum Zielprotein im Ca2+-gebundenen und eine geringere Bindungsstärke im Ca2+-freien Zustand aufweisen. Protein S aus M. xanthus wurde als scaffold zur Generierung künstlicher Bindeproteine verwendet die Ca2+-abhängig sfGFP binden. Durch die Randomisierung von 15 Aminosäurepositionen wurde eine Protein S Bibliothek erstellt und über Ribosomen-display Varianten mit bis zu sub-nanomolaren Affinitäten und selektiver Bindung erhalten. Es wurde gezeigt dass in Abwesenheit von Ca2+ nicht nur die Komplexbildung der Protein S-Variante und sfGFP unterbunden sondern auch bestehende Komplexe dissoziiert werden können. Eine Variante wurde auf einer Säulenmatrix immobilisiert und damit die Ca2+-abhängige chromatographische Reinigung von sfGFP aus Zellextrakten gezeigt. | |
In this work a new class of artificial binding proteins was developed which are based on Ca2+-sensitive scaffolds of the βγ-crystallin family. These new binding proteins act as molecular switches that show a high affinity to the target structure in the presence of Ca2+ and a low affinity in the Ca2+-free state. Protein S of M. xanthus was used as a scaffold for the generation of Ca2+-sensitive artificial binding proteins against sfGFP. By randomization of 15 positions a Protein S library was constructed and ribosome display was used for selection. Variants with affinities as low as sub-nM interaction could be isolated. It could be shown that the complex formation of Protein S and sfGFP could be prevented by the absence of Ca2+ and already formed complexes could be dissociated by titration of Ca2+ from the system. One of the Protein S variants was immobilized on the surface of a column matrix and the Ca2+-dependent purification of sfGFP from crude cell extracts was demonstrated. |
