In the present work an efficient easy-to-use screening method for the determination of vitamin D (ergosterol 7-dehydrocholesterol cholecalciferol ergocalciferol 25-hydroxycholecalciferol 25-hydroxyergocalciferol 1α 25-dihydroxycholecalciferol 1α 25-dihydroxyergocalciferol (24R)-24 25-dihydroxycholecalciferol) in food was developed in-house validated and applied. The suitability for routine analysis was proven by the study of more than 1 500 real samples. After spiking with isotope-labeled internal standards food samples were saponified purified and concentrated by combined solid-phase extractions (normal- and reversed-phase). Identification and quantification were performed by liquid chromatography-tandem mass spectrometry using scheduled multiple reaction monitoring mode (ESI positive).
