Um die Pathogenese der Sepsis und Endokarditis zu untersuchen wurde der Einfluss der Toxine α-Toxin von Staphylococcus aureus SLO von Streptococcus. pyogenes und LPS von E. coli auf den Aktivierungszustand von Thrombozyten und PMN untersucht. Dabei führte nur α-Toxin zu einer messbaren Aggregation am Aggregometer. Diese konnte durch Abciximab Iloprost NPN und Puromycin dosisabhängig inhibiert werden. Der Zusatz von PMN zu α-Toxin stimulierten Plättchen erhöhte die Aggregation signifikant. Des Weiteren wurde mittels Western Blot nachgewiesen dass SLO ebenso wie α-Toxin und das endogene Thrombin in der Lage sind die Bcl-3 Expression in Thrombozyten zu induzieren. Am Durchflusszytometer konnte die toxinvermittelte konzentrationsabhängige Aktivierung von Thrombozyten und PMN anhand verschiedener Aktivierungsparameter nachgewiesen werden. Dabei konnte durch den Zusatz von PMN in physiologischer Konzentration die toxinvermittelte Aktivierung von Thrombozyten signifikant erhöht werden. | |
To understand better the pathogenesis of sepsis and endocarditis we analyzed the influence of Staphylococcus aureus α-toxin Streptococcus. pyogenes SLO and E. coli LPS on the activation status of human platelets and PMN. We only examined an aggregation by α-toxin treated plate lets by aggregometry. Addition of PMN increased this aggregation rate significantly. By western blot analysis we could show thatα-toxin as well as SLO and endogenous thrombin can induce expression of Bcl-3 protein. By use of flow cytometry we determined toxin mediated activation of platelets and PMN that by use of different activation parameters. Also addition of PMN to α-toxin treated platelets increased significant platelet activation status. |
