Die vorliegende Arbeit liefert neue Einblicke in die komplexen Tumor-Stroma-Interaktionen im kolorektalen Karzinom (CRC): erstmals wurde gezeigt dass humane multipotente Stromazellen (MSC) des Knochenmarks spezifisch gegenüber anderen Stromazellen das Xenograftwachstum verschiedener CRC-Zelllinien fördern. Diese Wachstumsförderung manifestierte sich bereits in einer frühen Phase der Xenograftentwicklung und äußerte sich prominent in E-Cadherin dysfunktionalen CRC-Zellen. Im in vitro 3D-Modellwurde ermittelt dass E-Cadherin dysfunktionale CRC-Zellen und MSC über CRC- sowie MSC-exprimiertes β1-Integrin - vermutlich mit MSC-sezerniertem Kollagen I als Linker - interagieren wodurch der Aufbau einer Sphäroidstruktur vermittelt wird. In vivo wurde gezeigt dass die Interaktion über CRC-exprimiertes β1-Integrin spezifisch in α-Catenin defizienten CRC-Zellen für die MSC-vermittelte Förderung des Xenograftwachstums relevant ist und somit als mögliches Therapieziel dienen könnte. | |
This PhD thesis gives new insights into the complex tumor-stroma-interactions in colorectal carcinomas (CRC): Forthe first time it could be shown thathuman bone-marrow derived multipotent stromal cells (MSC)specifically compared to other stroma cells promote xenograft growth of various CRC-cell lines. This growth-promotion already appeared during an early stage of xenograft development and was prominent for E-cadherin dysfunctional CRC-cells. Within the in vitro 3D-model it was shown that E-cadherin dysfunctional CRC-cells and MSC interact through CRC- and MSC-expressed β1-integrin - probably with MSC-expressed collagen I as a linker - leading to the formation of a spheroid-structure. In vivo CRC-expressed β1-integrin is involved in MSC-mediated promotion of xenograft growth specifically in α-catenin deficient CRC-cells and therefore could be used as a potential target for a therapy. |
