Curcumin ein Inhaltsstoff aus Gelbwurz (Curcuma longa L.) ist seit einigen Jahren im Fokus der biomedizinischen Forschung. Neben seiner antioxidativen und antiinflammatorischen Wirkkomponente zeigt Curcumin auch vielversprechende Wirkungen bei der Therapie von Krebserkrankungen. Die meisten der beschriebenen Effekte wurden jedoch nur in in vitro Experimenten nachgewiesen und derzeit gibt es nur unzureichende Daten über die Wirkung von Curcumin in vivo. Ein Grund dafür ist die geringe orale Bioverfügbarkeit von Curcumin. Das Ziel dieser Arbeit war es eine Formulierung für die orale Applikation von Curcumin zu entwickeln. Die Substanz wurde dafür in Lipidnanopartikel eingearbeitet die mittels Hochdruckhomogenisation hergestellt wurden. Die hergestellten Formulierungen wurden hinsichtlich ihrer Partikelgröße und der physikochemischen Eigenschaften des Lipides und des inkorporierten Curcumins untersucht. Des Weiteren wurden ein in vitro Verdau-Assay und Freisetzungsversuche durchgeführt um zu simulieren wie sich die Partikel nach oraler Applikation verhalten. | |
The active ingredient of turmeric (Curcuma longa L.) Curcumin has been in the focus of biomedical research since many years. Besides its antioxidant and anti-inflammatory activity Curcumin has also shown promising potential against cancerous diseases. However many effects were only demonstrated by in vitro experiments and only few data of in vivo experiments exist. A major reason for the insufficient in vivo data is the low oral bioavailability of Curcumin. The aim of the present work was the development of a formulation for the oral delivery of Curcumin. The drug was incorporated into lipid nanoparticles which were produced by high pressure homogenization. The produced particles were characterized with regard to their particle size and to the physicochemical properties of the lipid and the incorporated Curcumin. Furthermore an in vitro digestion assay and release experiments were conducted to simulate the fate of the lipid and the drug during oral ingestion. |
