Zellsuspensionen von Eschscholzia californica reagieren auf den Kontakt mit einem Hefe-Elicitor mit der Bildung von Benzophenanthridin-Alkaloiden, welche als toxische Phytoalexine fungieren (Interkalation in doppelsträngige Nucleinsäuren, Störung der Membranpermeabilität, Hemmung von SH-abhängigen Enzymen). Die Toxin-produzierenden Zellen schützen sich a) durch Ausscheidung der Alkaloide b) durch Reduktion derjenigen Alkaloide, die aus dem Medium aufgenommen werden. Im Mittelpunkt der Arbeit standen Untersuchungen zur Metabolisierung von Benzophenanthridinen durch Zellsuspensionen von E. californica. Extern zugesetzte Benzophenanthridine, insbesondere Sanguinarin, werden durch intakte Zellen aufgenommen. Die Aufnahme ist eng mit der Reduktion dieser Alkaloide gekoppelt, so daß in den Zellen ausschließlich die gebildeten Dihydroalkaloide nachweisbar sind. Berberin, ein strukturverwandtes Isochinolinalkaloid, sowie Phenanthridin-Analoga (Ethidiumbromid, Propidiumjodid) werden weder aufgenommen noch umgesetzt. Das die Reduktion katalysierende Enzym, die Sanguinarin-Reduktase, wurde nach Zellaufschluß durch FPLC gereinigt. Das bisher unbekannte Enzym ist ein lösliches 30 kDa-Protein, welches die irreversible Reduktion von Sanguinarin mit NAD(P)H zu Dihydrosanguinarin katalysiert. Das Enzymprotein besitzt Sequenzähnlichkeiten mit einer 3 beta-Hydroxysteroid-Dehydrogenase/Isomerase aus Oryza sativa oder Arabidopsis thaliana. Typische Substrate dieser Enzymklasse werden durch die Sanguinarin-Reduktase jedoch nicht umgesetzt. Sehr wahrscheinlich unterliegt das gebildete und ausgeschiedene Sanguinarin einem Recycling, welches die Benutzung dieses Alkaloids als Phytoalexin erlaubt, gleichzeitig jedoch die Akkumulation des Toxins verhindert.

Zusammenfassung (Englisch)

Cultured cells of Eschscholzia californica respond to a yeast elicitor with overproduction of benzophenanthridine alkaloids that act as potent phytoalexins (due to intercalation into double stranded nucleic acids, interference with permeability of membranes, inhibition of SH-dependent enzymes). The toxin producing cells protect themselves a) by excreting the alkaloids b) by reduction of those alkaloids that are reabsorbed from the outer medium. The aim of the work was to analyse the metabolism of benzophenanthridine alkaloids by cultured cells of E. californica. External benzophenanthridine alkaloids, especially sanguinarine, are rapidly taken up by intact cells. The uptake is closely coupled with the reduction of the alkaloids with the result that in intact cells only dihydroalkaloids can be detected. The structurally related isoquinoline alkaloid berberine or phenanthridines as ethidium bromide and propidium iodide are neither taken up nor converted. The enzyme catalysing the reduction was named sanguinarine reductase and purified from the 15 000 g supernatant by FPLC. The newly found enzyme is a soluble 30 kDa protein catalysing the reduction of sanguinarine with NAD(P)H to yield dihydrosanguinarine. The enzyme protein contains sequences homologous to a 3 betahydroxysteroid dehydrogenase/isomerase from Oryza sativa or Arabidopsis thaliana. Typical substrates of this class of enzymes are not converted by the sanguinarine reductase. Most probably the sanguinarine built and excreted by the cells is subject to a recycling that allows the use of this alkaloid as a phytoalexin and at the same time prevents the accumulation of the toxin.
