Bei Prozessen wie Tumorwachstum, lokale Invasion, Intravasation, Extravasation, Angiogenese und Metastasierung spielen Ektoenzyme eine entscheidene Rolle. Aminopeptidase N (APN)/CD13 ist eine ubiquitär exprimierte, transmembrane Ektopeptidase, die im Tumorgeschehen mit Angiogenese, Metastasierung und Tumorinvasion in Zusammenhang gebracht wird. Autotaxin (ATX/NPP2) wurde ursprünglich als Motilitäts-steigerndes Protein beschrieben und besitzt neben PhosphodiesteraseI/ Nukleotidpyrophosphatase-Aktivität auch Lysophospholipase-D-Aktivität. Über die Expression, Regulation und Funktion von APN/CD13 und ATX beim Schilddrüsenkarzinom existierten zu Beginn der Arbeit keine Daten. Es wurde gezeigt, dass undifferenzierte anaplastische Schilddrüsenkarzinome mehr APN/CD13 bzw. ATX exprimieren als differenzierte Schilddrüsenkarzinome. Um die Funktion von APN/CD13 und ATX zu untersuchen, wurden stabile APN/CD13- bzw. ATX-überexprimierende Transfektanten der Zelllinie FTC-133 hergestellt. FTC-133-Zellen, die APN/CD13 oder ATX überexprimieren, zeigten eine erhöhte Zellmotilität und Veränderungen in der Genexpression des schilddrüsen-spezifischen Transkriptionsfaktors TTF-1, S100A3, Contactin-6, des Adhäsionsmoleküls Lumican und des "Cytokine-Receptor like"-Faktors CRLF. Es kann geschlussfolgert werden, dass APN/CD13 und ATX in einem mehrstufigen Prozess der Schilddrüsenkarzinogenese zu Dedifferenzierung und Tumorprogression beim Schilddrüsenkarzinom beitragen.

Zusammenfassung (Englisch)

Processes such as tumor growth, local invasion, intravasation, extravasation, angiogenesis and metastasis require the function of ectoenzymes. Aminopeptidase N (APN)/CD13 is a transmembrane ectopeptidase expressed on a wide variety of cells. In the area of cancer APN/CD13 is linked with angiogenesis, metastasis and tumor invasion. Autotaxin (ATX/NPP2) is a tumor cell motility-stimulating factor that displays both a nucleotide pyrophosphatase/ phosphodiesterase activity and a lysophospholipase D (lysoPLD) activity. However, the precise functions of APN/CD13 and ATX in tumor cells and the relationship of these enzymes to thyroid cancer has remained unclear. Undifferentiated anaplastic thyroid carcinomas expressed more APN/CD13 and ATX than differentiated thyroid carcinomas. To study the function of APN/CD13 and ATX in thyroid carcinoma cells, we stably transfected FTC-133 cells with an expression vector for APN/CD13 as well as for ATX. APN/CD13 or ATX overexpressing FTC-133 cells showed an increased motility and changes in differential gene expression of thyroid transcription factor (TTF)-1, S100A3, contactin-6, the cell adhesion molecule lumican and the cytokine-receptor like factor CRLF. In summary, both APN/CD13 and ATX was demonstrated to contribute to a multistep process of dedifferentiation and tumor progression in thyroid carcinomas.
