In dieser Arbeit wurden epigenetische Silencing Prozesse in Arabidopsisan transgenen LUCIFERASE-Repeats analysiert. Das Silencing derLUC-Repeats wird über heterochromatisches transkriptionelles Gen-Silencingzell erblich kontrolliert und durch H3K9me2 H3K27me1 DNA-Methylierung und nicht-kodierende Pol V-Transkripte vermittelt. Des Weiteren wurden durch Analysen im LUC-Testsystem isolierter Silencing-Suppressormutanten die Rolle bekannter und neuer epigenetischer Kontrollfaktoren imheterochromatischen Gen-Silencing untersucht. Die Identifizierung von Suppressormutanten mit unverändertem heterochromatischen Indexing an LUC-Repeats belegen dass heterochromatische Modifizierungen und PolV-Transkripte essentiell aber letztendlich nicht ausreichend für die Etablierung von heterochromatischem Gen-Silencing sind. Am Ende müssen für die finale Definierung von heterochromatischem Silencing noch bisher unbekannte epigenetische Prozesse stattfinden. Zur differenziellen Analyse von eu- und heterochromatisch lokalisierten Histonen wurde außerdem ein in vivo-Testsystem entwickelt.

In this work epigenetic silencing processes in Arabidopsis were analyzed at LUCIFERASE transgene repeats. Silencing of LUC repeats is controlled by heterochromatic transcriptional gene silencing involving H3K9me2 and H3K27me1 DNA methylation and non-coding Pol V dependent transcripts. Furthermore the role of known and new epigenetic factors in heterochromatic gene silencing was analyzed in isolated silencing suppressor mutants. Identification of silencing suppressor mutants with unchanged heterochromatic indexing of LUC repeats suggests that heterochromatic modifications and Pol V transcription are essential but not sufficient for heterochromatic gene silencing. For the final definition of heterochromatic gene silencing till now unknown epigenetic processes have to take place. In addition an in vivo test system was established for the differential analysis of euchromatic and heterochromatic localized histones.
