Der nuclear factor 90 (NF90) ist ein dsRNA-bindendes Protein und bildet einen Komplex mit nuclear factor 45 (NF45). Dieser Komplex übernimmt Funktionen im RNA-Metabolismus vor allem in post-transkriptionellen Genregulation. Für NF90-NF45 konnte auch gezeigt werden dass es Wirtsfaktor im Lebenszyklus einiger Viren z.B. Hepatitis C-Virus (HCV) ist. In der vorliegenden Arbeit erfolgte eine vergleichende biophysikalische Charakterisierung von NF90 / NF90 NF45 in vitro. Dabei wurde deutlich dass die Komplexbildung zur thermodynamischen Stabilisierung der Proteine Verbesserung der RNA-Bindung und Förderung einer neu-identifizierten RNA-Chaperon-Aktivität (RCA) von NF90 führt. Basierend auf der Interaktion von NF90-NF45 mit dem RNAGenom von HCV und anderen RNAs liefert diese Studie Details des Mechanismus der Reorganisation von RNA-Strukturen durch die RCA der Proteine. Folglich wird ein Modell aufgestellt in dem diese Aktivität mit der biologischen Funktion der Proteine korreliert. | |
The nuclear factor 90 (NF90) belongs to the dsRNA-binding protein family and forms a complex with the nuclear factor 45 (NF45). This complex is implicated to participate in RNA metabolism in particular in post-transcriptional gene regulation. NF90-NF45 is also involved in the life cycle of several viruses e.g. hepatitis C virus (HCV) which reflects its function as a host factor of viral amplification. This study reports a comparative biophysical characterization of NF90 and NF90-NF45 in vitro. Complex formation was demonstrated to result in a mutual thermodynamic stabilization of the proteins improved RNA binding by NF90 and facilitation of a newly identified RNA chaperone activity (RCA). Based on NF90-NF45’s interaction with the viral RNA genome of HCV and other RNA targets this study further presents detailed mechanistic insight on the rearrangement of RNA structures by the protein’s RCA. Hence a model is provided in which this activity correlates with its biological function. |
