Aktin reguliert die Lokalisation sowie die Aktivität von myocardin-related transcription factors (MRTFs) einer Kofaktor-Familie des serum response factors (SRF). Dabei verhindert die Bildung inhibitorischer G-Aktin:MRTF-A-Komplexe die Aktivierung der MRTF-SRF-abhängigen Transkription. WH2-Domänen aus Aktinbindeproteinen interagieren mit derselben Oberfläche des Aktinmonomers wie MRTF-A und könnten durch direkte Kompetition eine Komplexdissoziation stimulieren. In dieser Arbeit wird durch Bindungsexperimente belegt dass MRTF-A und isolierte WH2-Domänen-Varianten aus N-WASP WAVE2 JMY Cobl und Spire2 direkt um die G-Aktin-Bindung konkurrieren. Immunfluoreszenzstudien SRF-Reporterassays und qPCR-Analysen zeigten dass überexprimierte WH2-Domänen eine Kernakkumulation von MRTF-A und die MRTF-SRF-abhängige Genexpression induzieren. Diese WH2-vermittelte Aktivierung von MRTF-A konnte polymerisationsunabhängig erfolgen und findet vermutlich überwiegend im Zytoplasma statt. Somit liefert diese Arbeit neue Erkenntnisse für eine Regulation der G-Aktin:MRTF-A-Komplexdissoziation durch WH2-Domänen-Proteine. | |
Actin regulates the localization as well as the activity of myocardin-related transcription factors (MRTFs) a cofactor family of the serum response factor (SRF). The formation of inhibitory G-actin:MRTF-A complexes prevents the activation of MRTF-SRF-dependent transcription. WH2 domains from actin binding proteins interact with the same surface of the actin monomer as MRTF-A and may thus stimulate complex dissociation by direct competition. In this work binding experiments revealed that MRTF-A and isolated WH2 domain variants from N-WASP WAVE2 JMY Cobl and Spire2 compete directly for G-actin binding. Immunofluorescence studies SRF reporter assays and qPCR analysis showed that overexpressed WH2 domains induce a nuclear accumulation of MRTF-A and MRTF-SRF-dependent gene expression. This WH2-mediated activation of MRTF-A could occur polymerization-independent and predominantly takes place in the cytoplasm. Thus this work provides new insights for a regulation of G-actin:MRTF-A complex dissociation by WH2 domain proteins. |
