Die Genexpression hängt zu einem großen Teil von der zellulären Konzentration der mRNA ab, die wiederum von dem Verhältnis zwischen Transkription und Abbau bestimmt wird. Ein allgemeiner Weg des mRNA-Abbaus in Hefe und in höheren Eukaryonten wird durch eine allmähliche Verkürzung des Poly(A)-Schwanzes eingeleitet. Die Deadenylierung ist ein relativ langsamer Proezss und wahrscheinlich geschwindigkeitsbestimmend für den mRNA-Abbau. Die poly(A)-spezifische Ribonuclease (PARN) ist eine 3'-5'-Exoribonuklease, die aus Kalbsthymus, HeLa-Zellen und Xenopus laevis gereinigt worden ist und vermutlich an der Deadenylierung der mRNA beteiligt ist. Diese Arbeit zeigt, daß die Deadenylierung in Extrakten aus HeLa-Zellen oder durch die gereinigte PARN durch ein m7GpppG-cap am RNA-Substrat stimuliert wird. PARN selber ist ein cap-bindendes Protein, was durch seine Fähigkeit, an m7GTP-Sepharose zu binden, gezeigt wurde. Die menschliche PARN wurde in HEK293-Zellen transient exprimiert und zur Homogenität gereinigt. Dieses Enyzme baute die Poly(A)-Schwänze von RNA mit einem m7GpppG-cap auf prozessive Weise ab, während RNA mit einem unmethylierten cap auf distributive Weise abgebaut wurde. Möglicherweise induziert die Bindung von PARN an das cap eine Konformationsänderung des Enzyms. Die Zugänglichkeit des 5'-cap könnte durch den Status des Translationsinitiations-Komplexes beeinflußt werden, der unter anderem aus dem cap-bindenden Protein eIF4E, eIF4G und dem Poly(A)-Bindungsprotein PABPC besteht. PARN wurde durch eIF4E nicht beeinflußt, aber durch eIF4G gehemmt. Eine Wechselwirkung zwischen PARN und der mittleren Domäne von eIF4G wurde aufgrund von GST- und m7GTP-pulldown-Experimenten vermutet. Das native Molekulargewicht von PARN wurde als 150 kDa bestimmt, während die monomere Masse 74 kDa beträgt. PARN scheint also als Homodimer zu existieren.

Gene expression depends to a large extent on the concentration of mRNA in the cell, which in turn is determined by the ratio between transcription and degradation. A general mRNA-degradation pathway in both yeast and higher eukaryotes is initiated by a gradual shortening of the poly(A) tail. Deadenylation is a relatively slow process and likely to be the rate-limiting step in mRNA turnover. The poly(A)-specific ribonuclease PARN is a 3' to 5' exoribonuclease that has been purified from calf thymus, HeLa cells and Xenopus laevis and is likely involved in mRNA deadenylation. This study shows that deadenylation in HeLa cell extracts and by the purified bovine PARN is stimulated by an m7GpppG-cap on the RNA substrates. Furthermore, PARN itself is a cap-binding protein, which is demonstrated by its ability to bind to m7GTP-Sepharose. Recombinant human PARN that was transiently expressed in HEK293 cells and purified to near homogeneity degraded poly(A) tails of m7GpppG-capped RNAs in a processive mode and RNAs with an unmethylated cap in a distributive mode of action. Therefore, binding of PARN to a bona fide cap may induce a conformational change of the enzyme. The accessibility to the 5'-cap might be influenced by the status of the translation initiation complex, containing, among others, the cap-binding protein eIF4E, eIF4G and the poly(A)-binding protein PABPC. PARN activity was not affected by eIF4E alone but was inhibited by eIF4G. A physical interaction between PARN and the middle domain of eIF4G (MIF4G) was suggested by GST- and m7GTP-pulldown assays. The native molecular mass of PARN was determined to be 150 kDa, whereas the monomeric molecular mass is 74 kDa. This suggests that PARN exists as a homodimer.
